Sue Perkins has two siblings; David Perkins and Michelle Perkins. They are her younger siblings.

Growing up with her two younger siblings in Croydon, Susan’s father worked at a local car dealership, and her mother served as a secretary. She received her education at Croham Hurst School, an independent girls’ school in South Croydon. During her time at school, she crossed paths with television presenter Susanna Reid.

After school, Susan pursued an education in English at the University of Cambridge, specifically at New Hall (now Murray Edwards College), where she graduated in 1991. Her association with the Footlights, a renowned Cambridge comedy club, led her to meet her creative partner, Mel Giedroyc. Susan served as the Footlights president during the academic year 1990–91.
